What happened?
BioMarin has agreed to acquire Alesta Therapeutics to gain ALE1, an oral small-molecule therapy being developed for hypophosphatasia, or HPP. The deal adds a clinical-stage programme to BioMarin’s pipeline and is expected to close during the third quarter, subject to customary conditions.
Why does it matter?
ALE1 could become the first oral therapy for HPP, offering a potential alternative to the injectable treatments currently available. The candidate is designed to address excess inorganic pyrophosphate, a central disease metabolite, with the aim of improving bone and mineral metabolism across the wider spectrum of HPP manifestations.
The transaction also gives BioMarin an opportunity to expand into a larger rare disease market while strengthening its early-stage clinical pipeline.
Who’s behind it?
BioMarin is a rare disease biotechnology company with commercial therapies and a pipeline focused on genetically defined conditions. Alesta is developing oral small-molecule treatments for underserved diseases, with ALE1 as its lead programme.
How was it achieved?
ALE1 is currently being evaluated in an ongoing Phase 1/2a trial in healthy volunteers and adults with HPP. The study is assessing safety, tolerability, pharmacokinetics and pharmacodynamics.
Under the agreement, Alesta will spin out all non-ALE1 assets into a new entity before closing. Its employees will transfer to that entity, so no Alesta staff will join BioMarin through the acquisition.
What’s next?
BioMarin will take responsibility for advancing ALE1 after the transaction closes. The company plans to provide updated full-year 2026 financial guidance following completion and expects the deal, excluding the upfront payment, to have a modest dilutive effect on 2026 results.
